SARS-CoV-2 escapes direct NK cell killing through Nsp1-mediated downregulation of ligands for NKG2D
Abstract
Summary Natural killer (NK) cells are cytotoxic effector cells that target and lyse virally-infected cells; many viruses therefore encode mechanisms to escape such NK cell killing. Here, we interrogated the ability of SARS-CoV-2 to modulate NK cell recognition and lysis of infected cells. We found that NK cells exhibit poor cytotoxic responses against SARS-CoV-2-infected targets, preferentially killing uninfected bystander cells. We demonstrate that this escape is driven by downregulation of ligands for the activating receptor NKG2D (“NKG2D-L”). Indeed, early in viral infection, prior to NKG2D-L downregulation, NK cells are able to target and kill infected cells; however, this ability is lost as viral proteins are expressed. Finally, we found that SARS-CoV-2 non-structural protein 1 (Nsp1) mediates downregulation of NKG2D-L and that Nsp1 alone is sufficient to confer resistance to NK cell killing. Collectively, our work reveals that SARS-CoV-2 evades NK cell cytotoxicity and describes a mechanism by which this occurs.

Incredible. Sweden does the conservative thing, the kind of thing recommended by the WHO for decades, and it is called "radical". Meanwhile, what China and then Italy did (to its shame) is regarded as the new normal way of doing things. The kind of people behind this new authoritarian, technocratic ideology have no qualms about weakening and disrupting western societies. Why? So they can 'build back better' of course! Sure, it may mean that inflation is at 10 percent and interest rates are steadily rising, and the world's poor are facing starvation in increasing numbers, and family farmland is being legislated out of operation, but just bear with it for a while and it will be worth it! Actually we have not been in contact after the tests of the UNIBO report, but I always considered and continue to consider him an intelletually honest individual. He is a Professor of Physics of one of the most important universities of the world. Everybody has understood that he has been forced to sign the “erratum” and I can understand why he signed it: he has two daughters and one son, all aged between 12 and 18 years…
He would never write that pout-purri of stupidities, written from somebody that has a very poor education in the matter, let alone sign them in a normal situation. He can’t talk, but we can understand all the same.
