    Human naïve antibodies contain anti-SARS-CoV-2 RBD antibodies

    The virus elicits neutralizing antibodies that prevent viral entry into the host cells. Most of these antibodies are directed against the viral spike glycoprotein, specifically its receptor-binding domain (RBD), a small immunodominant region. Antibodies to the RBD are of several types, each type targeting a different epitope, though there is a degree of overlap. This indicates that neutralization can occur via multiple mechanisms occurring via inhibition of viral entry at more than one site on the RBD.


    The majority of RBD antibodies described so far are surprisingly similar to their germline precursors, a state called low somatic hypermutation. This means that not only is it easy to produce anti-SARS-CoV-2 RBD antibodies, it makes it convenient for researchers to use naïve antibody libraries to pick out novel therapeutic antibodies and to compare antibody clones selected by their ability to bind the viral antigens in vitro with those that arise during natural infection or vaccination.


    The researchers used yeast display techniques to isolate three novel neutralizing antibodies from one such library. Of these antibodies, ABP18 blocks RBD-ACE2 binding, while two (NBP10 and NBP11) do not. Thus, the technique proved to be capable of differentiating antibodies that blocked ACE2 and those that did not.

    A shadow over the promising inhaled interferon beta COVID-19 therapy has been cleared with the discovery that although it appears to increase levels of ACE2 protein - coronavirus' key entry point into nose and lung cells - it predominantly increases levels of a short version of that protein, which the virus cannot bind to.

    Naturally occurring antiviral proteins called interferons have shown promise in treating COVID-19. However, previous studies have shown that interferons increase levels of ACE2 - casting doubts over the potential for such treatments, with the possibility that increased ACE2 could see these drugs actually worsen COVID-19 impacts.


    But this latest research shows that it is predominantly the short ACE2, which lacks the viral binding site, that is increased in response to interferons. Since levels of the longer form of ACE2 remain unchanged, interferons do not appear to boost entry points for the virus, supporting their use in treating COVID-19 patients.

    The genetic mutations in new strains of the coronavirus could affect the results of some COVID-19 tests, according to a warning from the Food and Drug Administration (FDA). Because the mutations affect the part of the virus's genetic material that these tests look for, the tests could end up giving a false negative result, meaning the results are negative but an infection is present.


    The FDA recently discovered that three particular COVID-19 tests could give a false negative result if the sample contains the B.1.1.7 variant of the virus, which triggered the London lockdown in December. It has since spread to other countries, including the U.S. The risk for a false negative in this situation is low, according to the letter. But the FDA is flagging the issue to health care workers and lab staff.

    While researching new studies around the globe, I came across this interesting piece of information on Nature.com that was published in November. Nature carries a compilation of research from around the world and this one research said scientists at the Francis Crick Institute in London analysed blood samples—from adults and children—who had not been infected with the new virus and found they contained antibodies. Nature says: “Scientists have found antibodies that recognize SARS-CoV-2 in the blood of people who have never caught the virus. Children are particularly likely to harbour such antibodies, which might explain why most infected children have either mild illness or none at all.” The blood samples in question were collected before Covid began to wreak havoc worldwide or just when it started to, which means that immunity to it already existed in some people.

    Not exactly: It's (How CoV-19 virus has been created inside the Wuhan lab) based on the opinion of some of the most famous experts in the field of genetics. A random mutation path form all known paths, Pangolin etc. virus to SARS-CoV-19 would take at least 20 years. Not even to talk that this (the best fitting ) virus had to walk some 1000 miles and must exactly land in a human with the right condition. May be a Pangolin on drugs can do this..walking 1000 miles ( if It manages to escape 100'000'000 Chines hunters..)

    Further the same work has already been done at the Wuhan-Lab once before with an influenza virus... For details of the experiment please ask Fauci! He paid for it = did authorize the payment!

    Abstract

    The coronavirus disease 19 (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has affected millions of people worldwide. Recent evidence raised the question about the possibility that cats may be a domestic host for SARS-CoV-2 with unknown implications in disease dissemination. Based on the fact that the domestic cat flea, Ctenocephalides felis, are abundant ectoparasites infesting humans, companion animals and wildlife and that coronavirus-like agents have been identified in the ectoparasite tick vector, Ixodes uriae of seabirds, herein we considered the presence of coronaviruses in general and SARS-CoV-2 in particular in C. felis. We identified coronavirus-derived and cell receptor angiotensin-converting enzyme RNA/proteins in C. felis. Although current evidence suggests that pets are probably dead-end-hosts with small risk of transmission to humans, our results suggested that cat flea may act as biological and/or mechanical vectors of SARS-CoV. Although preliminary, these results indicate a possibility of ectoparasites acting as reservoirs and vectors of SARS-CoV and related beta-coronavirus although with little disease risk due to systemic transmission route, low viremia, virus attenuation or other unknown factors. These results support the need to further study the role of animal SARS-CoV-2 hosts and their ectoparasite vectors in COVID-19 disease spread.

  • Some weeks ago the international FM/R/J press started a concentrated effort to blame Sweden for the bad tactics not to follow the international force an fear behavior.


    "Sweden has to put in place more strong measures" was the key spell spread.


    But for people following the Sweden data it was clear that only the case numbers did increase but the number of deaths all the way did go down. Today these are 3x lower that in Switzerland and 5x lower than in Germany despite higher case numbers...


    So what went wrong - outside Sweden??

    Sinovac: Brazil results show Chinese vaccine 50.4% effective

    A coronavirus vaccine developed by China's Sinovac has been found to be 50.4% effective in Brazilian clinical trials, according to the latest results released by researchers.


    It shows the vaccine is significantly less effective than previous data suggested - barely over the 50% needed for regulatory approval.


    The Chinese vaccine is one of two that the Brazilian government has lined up.


    Brazil has been one of the countries worst affected by Covid-19.


    Sinovac, a Beijing-based biopharmaceutical company, is behind CoronaVac, an inactivated vaccine. It works by using killed viral particles to expose the body's immune system to the virus without risking a serious disease response.


    Several countries, including Indonesia, Turkey and Singapore, have placed orders for the vaccine.

The drug is targeted toward people who are recently diagnosed with Covid-19, with the hopes of preventing the need for hospitalization. Regeneron's Covid-19 antibody treatment, which President Donald Trump received after contracting the disease, also has received limited clearance from the FDA.
