    Penn Researchers Discover Drug that Blocks Multiple SARS-CoV-2 Variants in Mice

    A small molecule STING agonist was highly protective against the virus that causes COVID-19 and likely other coronaviruses

    PHILADELPHIA – The drug diABZI — which activates the body’s innate immune response — was highly effective in preventing severe COVID-19 in mice that were infected with SARS-CoV-2, according to scientists in the Perelman School of Medicine at the University of Pennsylvania. The findings, published this month in Science Immunology, suggest that diABZI could also treat other respiratory coronaviruses.


    “Few drugs have been identified as game-changers in blocking SARS-CoV-2 infection. This paper is the first to show that activating an early immune response therapeutically with a single dose is a promising strategy for controlling the virus, including the South African variant B.1.351, which has led to worldwide concern,” said senior author Sara Cherry, PhD, a professor of Pathology and Laboratory Medicine and scientific director of the High-Throughput Screening (HTS) Core at Penn Medicine. “The development of effective antivirals is urgently needed for controlling SARS-CoV-2 infection and disease, especially as dangerous variants of the virus continue to emerge.”


    The SARS-CoV-2 virus initially targets epithelial cells in the respiratory tract. As the first line of defense against infection, the respiratory tract’s innate immune system recognizes viral pathogens by detecting their molecular patterns. Cherry and her research team first sought to better understand this effect by observing human lung cell lines under the microscope that had been infected with SARS-CoV-2. They found that the virus is able to hide, delaying the immune system’s early recognition and response. The researchers predicted that they may be able to identify drugs — or small molecules with drug-like properties — that could set off this immune response in the respiratory cells earlier and prevent severe SARS-CoV-2 infection.


    To identify antiviral agonists that would block SARS-CoV-2 infection, the researchers performed high throughput screening of 75 drugs that target sensing pathways in lung cells. They examined their effects on viral infection under microscopy and identified nine candidates —including two cyclic dinucleotides (CDNs) — that significantly suppressed infection by activating STING (the simulation of interferon genes).


    Since CDNs have low potency and make poor drugs, according to Cherry, she and her team decided to also test a newly-developed small molecule STING agonist called diABZI, which is not approved by the Food and Drug Administration but is currently being tested in clinical trials to treat some cancers. The researchers found that diABZI potently inhibits SARS-CoV-2 infection of diverse strains, including variant of concern B.1.351, by stimulating interferon signaling.


    Finally, the researchers tested the effectiveness of diABZI in transgenic mice that had been infected with SARS-CoV-2. Because the drug needed to reach the lungs, diABZI was administered through a nasal delivery. diABZI-treated mice showed much less weight loss than the control mice, had significantly-reduced viral loads in their lungs and nostrils, and increased cytokine production — all supporting the finding that diABZI stimulates interferon for protective immunity.


    Cherry said that the study’s findings offer promise that diABZI could be an effective treatment for SARS-CoV-2 that could prevent severe COVID-19 symptoms and the spread of infection. Additionally, since diABZI has been shown to inhibit human parainfluenza virus and rhinovirus replication in cultured cells, the STING agonist may be more broadly effective against other respiratory viruses.


    “We are now testing this STING agonist against many other viruses,” Cherry said. “It’s really important to remember that SARS-CoV-2 is not going to be the last coronavirus that we will see and will need protection against.”

    No attendees at an indoor concert that employed rapid COVID-19 lateral-flow screening, N95 respirators, and a well-ventilated venue tested positive for COVID-19 in the next 8 days, showing no increased virus transmission risk associated with the event, according to preliminary findings from a randomized, controlled trial published yesterday in The Lancet Infectious Diseases.


    A team led by researchers from the Germans Trias i Pujol University Hospital (GTPUH) in Barcelona, Spain, compared the coronavirus infection rate in 465 concert-goers aged 18 to 59 who were screened for COVID-19 and fever before entry and wore an N95 respirator throughout the event with 495 participants who were screened and then asked to go home. No concert-goers or 58 staff members tested positive 8 days after the event, compared with 2 in the control group.


    "Our study provides early evidence that indoor music events can take place without raising the risk of SARS-CoV-2 transmission when comprehensive safety measures are in place, but it is important that our findings are considered in light of the situation in Spain at the time—when cases were not high and many restrictions were in place, senior study author Josep Llibre, MD, PhD, of GTPUH, said in a Lancet news release emailed to journalists.


    "As a result, our study does not necessarily mean that all mass events are safe."


    All venue entry, exit doors open

    The concert took place in Barcelona on Dec 12, 2020, at a time when the prevalence of COVID-19 infections was low to intermediate, at 221 per 100,000 people, local travel was restricted, indoor gatherings were limited to six people, and COVID-19 vaccines had not yet been rolled out.


    In a post-event questionnaire, concert-goers reported a satisfaction score of, on average, 8.6 out of 10 when asked to rate how well they were able to enjoy the performance. When asked how willing they would be to attend another event with the same mitigation protocols, the average score was more than 9 out of 10 (10 being most willing).


    All participants were required to install two smartphone apps, one for contact tracing and one to receive their COVID-19 test results, complete health questionnaires before and 10 days after the event, and fill out a satisfaction survey.


    Most concert-goers spent about 2 hours and 40 minutes inside the venue during the 5-hour event. In addition to two live music acts, there were two DJ performances. Drinks, including alcohol, were available in a separate room, and attendees could smoke outside in a limited-capacity area with physical distancing.


    Concert attendees weren't asked to physical distance and were allowed to sing and dance in the concert hall, although the cloakroom was closed to prevent lines from forming. All entry and exit doors remained open throughout the event, and the venue was temperature controlled to make wearing an N95 respirator more comfortable.


    Before the event, 28 people tested positive for COVID-19 on a transcription-mediated amplification test, 13 in the event group and 15 controls. Further lab tests confirmed that none were infected with live virus, indicating they were unlikely to spread COVID-19. All 28 had been diagnosed as having coronavirus about 7 weeks before the concert.


    Future lines of research

    Llibre cautioned that the strict protocols used in the study may be difficult to replicate or cost-prohibitive in other venues or in larger gatherings and that future research is needed to understand mass gatherings in other contexts of the pandemic.


    "Widespread vaccination campaigns, changes in local incidence and the emergence of SARS-CoV-2 variants with higher transmissibility could all impact the interventions we tested, so we need more studies including larger numbers of people that explore different scenarios and policies that take into consideration the local context," he said.


    First author Boris Revollo, MD, also of GTPUH, said in the release that while large gatherings are tied to a high risk of SARS-CoV-2 transmission, cancellation of these events has resulted in significant financial losses. "As societies look toward the possibility of safely resuming cultural activities, lateral flow tests, which can deliver results within 30 minutes and be taken on site, have been proposed as a means of screening people on entry to enable large events to take place," he said.


    In a commentary in the same journal, Rosanna Peeling, PhD, and David Heymann, MD, of the London School of Hygiene and Tropical Medicine, said that the study is an important reminder of the need for more studies as diagnostic test innovation continues.


    Such research could answer questions, they said, such as, "Would triple-layered masks have been sufficient? How does rapid antigen testing at entry compare with molecular screening within 72 h of entry? Does screening create bottlenecks with an increased risk of transmission among those not yet tested? … And the question of concern for all is whether existing [rapid diagnostic tests] are able to detect COVID-19 variants of concern and the role of vaccination."

    It sounds a little hard to swallow, but a new study suggests that how people react to bitter flavors correlates with the severity of their COVID-19 infection.


    It’s an exciting insight because during the past 16 months, it’s become clear that people don’t respond to SARS-CoV-2 predictably. It’s been impossible to determine whether someone will experience mild symptoms or develop life-threatening respiratory disease. Imagine if a simple taste test could indicate a person’s risk of developing severe COVID-19.


    Henry Barham, a rhinologist at the Baton Rouge General Medical Center, in Louisiana, published a study in the medical journal JAMA Network Open on May 25 that analyzed nearly 2,000 patients and found that “supertasters”—individuals who are overly sensitive to some bitter compounds—were less likely to test positive for the virus. If this association holds true, it implies, for example, that people who don’t find broccoli too bitter are in a higher risk group for severe COVID-19.


    “This is a very interesting study that suggests that receptors on our tongue that allow us to sense bitter flavors are also linked to our vulnerability to respiratory infections like COVID-19,” says David Aronoff, director of the division of infectious diseases at Vanderbilt University Medical Center, in Nashville, Tennessee, who was not involved with this research. That taste receptors may also be involved with immunity is surprising, he says.


    Do “supertasters” have super powers?

    At Yale University in the 1990s, psychologist Linda Bartoshuk pioneered the study of genetic variations in taste perception. She coined the term “supertaster” to describe the 25 percent of people who are intensively sensitive to bitter flavors. Another 25 percent of people are “nontasters” who barely detect bitter flavors, and the remaining 50 percent are just “tasters”—those who register the bitterness but not to the degree that it’s distasteful.



    Supertasters are more sensitive to bitter flavors because they have up to four times as many taste buds on their tongue. Bitter compounds in certain foods and drinks are recognized by type 2 taste receptors, which are made by a family of genes called T2R. The T2R38 gene is among the best studied of these. Variations in the structure of the T2R38 protein the gene encodes correlates with a person’s tolerance of bitter compounds—such as phenylthiocarbamide and propylthiouracil—which are abundant in many vegetables, including broccoli, cabbage, and brussels sprouts.


    This is not the first time being a supertaster had been linked to a medical condition. Supertasters have a higher likelihood of having polyps in their colon, a risk factor for cancer associated with lower intake of those bitter vegetables.


    But supertasters can experience physiological advantages as well. As it happens, the T2R38 proteins are found in places other than the tongue. These “extraoral” areas include the epithelial cells that line the nose and upper respiratory tract, where they respond to invading pathogens.


    A 2012 study led by Noam Cohen, a rhinologist at the University of Pennsylvania, in Philadelphia, found that bacteria responsible for sinus infections activate the T2R38 protein receptors on cells lining the respiratory tract, causing them to produce nitric oxide. Nitric oxide is a key component of our immune response, the first line of defense against invading pathogens. It stimulates hair-like structures called cilia in the respiratory passageways that remove foreign particles and pathogens from the body. Consequently, supertasters experience fewer bacterial sinus infections.


    Barham, who studies T2Rs as they relate to innate immunity within the respiratory tract, was also aware that nitric oxide could poison SARS-CoV, a coronavirus (related to SARS-CoV-2 that causes COVID-19) first reported in Asia in 2003 that caused a respiratory illness that spread to 22 countries before it was contained. This prompted him to investigate whether a link exists between COVID-19 and supertasters

    I posted a video of Dr John Campbell in January, he advised to aspirate before vaccinate. I asked about this when I got my jab and was told that they don't do that anymore, it's the old fashion way!

    Amid calls for a fresh probe into the origins of Covid-19, an explosive new

    study has found that Chinese scientists created the virus in a lab in Wuhan, then tried to cover their tracks by reverse-engineering versions of the virus to make it look like it evolved naturally from bats.

    The novel coronavirus SARS-CoV-2 virus has no "credible natural ancestor" and was created by Chinese scientists who were working on a 'Gain of Function' project in a Wuhan lab, the Daily Mail reported on Sunday, citing a new research paper by British professor Angus Dalgleish and Norwegian scientist Dr Birger Sorensen.

    The new research claims that scientists took a natural coronavirus "backbone" found in Chinese cave bats and spliced onto it a new "spike", turning it into the deadly and highly transmissible Covid-19.

    The paper also quotes that researchers found "unique fingerprints" in Covid-19 samples that they say could only have arisen from manipulation in a laboratory.




    Authors Dalgleish and Sorensen wrote in their paper that they had prima facie evidence of retro-engineering in China' for a year, but were ignored by academics and major journals, reported DailyMail.com.

    The study alleged "deliberate destruction, concealment or contamination of data" at Chinese labs and notes the silencing and disappearance of scientists in China who spoke out about the activities.

    The research, has been obtained by DailyMail.com, is expected to intensify the ongoing debate on China's role in creating the virus that has claimed thousands of lives.

    In the 22-page paper which is set to be published in the scientific journal Quarterly Review of Biophysics Discovery, Dalgleish and Sorensen pieced together how Chinese scientists, some working with American universities, allegedly built the tools to create the coronavirus.

    Incidentally, 'Gain of Function' projects, which involves tweaking natural viruses to make them more infectious, had been outlawed by former US President Barack Obama.

    In an exclusive interview with DailyMail.com, Sorensen said that four amino acids on the spike have a positive charge, which causes the virus to tightly cling to the negatively charged parts of a human, becoming more infectious.

    Because these positively charged amino acids also repeal each other, it was rare to find even three in a row in naturally occurring organisms, while four in a row is 'extremely unlikely, he said.

    "The laws of physics mean that you cannot have four positively charged amino acids in a row. The only way you can get this is if you artificially manufacture it," Dalgleish told DailyMail.com.

    "The implication of our historical reconstruction, we posit now beyond reasonable doubt, of the purposively manipulated chimeric virus SARS-CoV-2 makes it imperative to reconsider what types of Gain of Function experiments it is morally acceptable to undertake," they wrote.

    In the paper, the scientists wrote that a natural virus pandemic would be expected to mutate gradually and become infectious but less pathogenic, which did not happen in the case of Covid-19.

    Dalgleish and Sorensen claimed that after the pandemic began, Chinese scientists took samples of the COVID-19 virus and 'retro-engineered' it, making it appear as if it had evolved naturally.

    "We think that there have been retro-engineered viruses created. They've changed the virus, then tried to make out it was in a sequence years ago," Dalgleish told DailyMail.com.

    The scientists also highlighted that Chinese scientists who wished to share their knowledge have not been able to do so or have disappeared.

    Sorensen said that he believes the virus escaped from lower security areas of the institute, where he believes Gain of Function research was performed, reported DailyMail.com.

    "We have seen lab leaks and we know it's happening. We also know from the reports we've seen, that coronavirus is worked on in Biosafety Level 2 or 3 labs. If they do Gain of Function in such labs, what do you expect?" he said.

    In February 2020, Botao Xiao, a molecular biomechanics researcher at South China University of Technology, published a paper claiming "the killer coronavirus probably originated from a laboratory in Wuhan," highlighting safety issues at the institute. However, he withdrew the paper weeks later after Chinese authorities denied any accidents at the lab.

    Dalgleish further told DailyMail.com that he believed China's resistance to the theory that Covid-19 is a man-made, escaped virus comes from scientists fearful that the revelation would shut down their field.

    "This looks like a weak defence to protect the discipline so that this type of genetic engineering will not be interfered with. I make no bones about it. The Gain of Function engineering should have been banned ages ago," he said.

    Despite former MI6 chief Sir Richard Dearlove saying publicly that the scientists' theory should be investigated, they said their research was dismissed by major scientific journals that were adamant that the virus jumped naturally from bats, reported DailyMail.com.

    However, leading academics, politicians and the media have now begun to contemplate the possibility that Covid-19 escaped from the Wuhan Institute of Virology (WIV) in China - where experiments included manipulating viruses to increase their infectiousness in order to study their potential effects on humans.

    Meanwhile, US President Joe Biden on May 26 said he has asked the US intelligence community to "redouble their efforts" to come to a conclusion on the origins of Covid-19 and report back to him within 90 days.

    China state media rejected the idea that Covid-19 had originated there and said that it is "a conspiracy created by US intelligence agencies"

    A few days ago, an article was published in the Wall Street Journal which revealed that three researchers at Wuhan Institute fell ill in November 2019 and had to be hospitalized. The report was based on previously undisclosed US intelligence. The researchers were admitted into the hospital a month before China reported the first case of Covid-19.

    The revelation has sparked debate and raised questions about whether China knew of the deadly virus way before it apprised the world.

    Recently, former New York Times science journalist Nicholas Wade published an article in the Bulletin of the Atomic Scientists where he argues that evidence is stronger than the virus leaked from a lab than that it occurred naturally.

    In an elaborate report, Wade said that much of the work of Chinese virologists on gain-of-function in coronaviruses was performed at the BSL2 safety level lab, which requires taking fairly minimal safety precautions. The pandemic broke out in the Chinese city of Wuhan in December 2019."For the lab escape scenario, a Wuhan origin for the virus is a no-brainer.

    Before that, an Australian daily revealed that Chinese scientists were thinking about bioweapons, visualising a World War-III scenario. The Weekend Australian newspaper cited a Chinese government document that discussed the weaponisation of SARS coronavirus.

    DailyMail.com further reported that China had installed the first of a planned five to seven Biolabs in Wuhan in 2017 for the purpose of studying the most high-risk pathogens, including the Ebola and the SARS viruses.

    In March, World Health Organization (WHO) Director-General Tedros Ghebreyesus also called for further studies after the release of an inconclusive report on an international team's field visit to Wuhan to research the origins of Covid-19, citing difficulties accessing raw data.
