    Obviously has weaknesses due to the nature of the self reporting of results, interpretation of results, and all the other usual research issues.


    Research suggests factors for risk and protection from Covid-19


    A new paper in Biology Methods & Protocols, published by Oxford University Press, investigated risk factors and protections against contracting COVID-19, and also for suffering from severe COVID. Researchers identified several characteristics – including male sex, lower age, blood group B, and larger household size – that increased the risk of infection and many other characteristics – including mask-wearing and using vitamin D – that decreased it. They also identified the things that increased or decreased the risk of a severe course of COVID-19. Some of these findings are unexpected.


    Researchers here ran a study on 30 thousand internet users, recruiting participants using sites like Facebook and Twitter, in the Czech Republic who shared information about their exposure to 105 risk factors for COVID-19.


    Many predictable behaviors appeared to protect people against the infection, while three factors, namely being actively involved in sports, frequent singing, and cold-water swimming, increased the risk of infection. The most substantial protective factor against COVID-19 infection was strict adherence to mask-wearing. The second most substantial protective factor was the consumption of vitamins and supplements. This study found in particular that taking vitamin D provided significant protection against acquiring COVID-19.


    Adherence to social distancing and frequent handwashing had only a weak protective effect. This study also indicated that having type B blood increased the risk for COVID-19 infection. Having cats or dogs as pets did not affect the risk of infection and had a mostly insignificant positive effect on the risk of someone experiencing a severe course of the disease.


    The study here found that tobacco smoking and also marijuana use (in women) have a relatively strong protective effect against infection. The researchers found that marijuana use and tobacco smoking could also have some protective effects against a severe course of COVID-19. The protective effects of tobacco smoking against infection have been reported in previous studies. However, most studies show adverse effects of smoking on the risk of a severe course of COVID-19.


    The most unexpected result of the study was a positive correlation between higher severity of the course of COVID-19 and adherence to wearing masks and respirators and, to a lesser extent, keeping social distance. The investigators speculate this is because people predisposed to a severe course of COVID-19 (those who are overweight, suffer from chronic obstructive pulmonary disease, or have diabetes) tried hard to avoid infection and more strictly adhered to public health recommendations about masks and social distancing. But when they did become infected, they often had a more severe course of the disease due to their underlying health problems.

Chiu et al published a report where both cardiac symptoms and ECG changes were recorded after the first and second injections. The results are alarming. After the second injection of mRNA 17.1% of students reported cardiovascular symptoms.

    Abstract

    Vitamin D deficiency has been associated with reduced neurocognitive functioning and the neurodegenerative processes. However, existing evidence on brain structural correlates of vitamin D deficiency is controversial. We sought to investigate associations of vitamin D levels with imaging patterns of brain aging. In addition, we investigated whether low vitamin D levels were associated with gray matter volumes, whole brain volumes and hippocampus volumes. Structural MRI data and vitamin D levels were obtained in 1,865 subjects from the general population. Linear regressions were applied to investigate the association of vitamin D levels and vitamin D deficiency with imaging derived brain age, total brain, gray matter and hippocampal volumes. Different sets of covariates were included. Vitamin D deficiency was significantly associated with increased brain age. Also, linear vitamin D levels were significantly associated with total brain and gray matter volumes, while no significant association with hippocampal volume was found. Further interaction analyses showed that this association was only significant for male subjects. Our results support previous findings suggesting that vitamin D-deficient individuals have an accelerated brain aging. In addition, associations between vitamin D levels and total brain/ gray matter volumes suggest neuroprotective effects of vitamin D on the brain.

    Fewer cases of melanoma were observed among regular users of vitamin D supplements than among non-users, according to a new study from the University of Eastern Finland and Kuopio University Hospital. The study findings have been published in Melanoma Research.


    Vitamin D is vital for the normal function of the human body, and it may also play a role in many diseases. The link between vitamin D and skin cancers has been extensively studied, but the studies have mainly focused on serum levels of calcidiol, which is a metabolite of vitamin D, and its association with skin cancers. The findings from these studies have been inconclusive and contradictory, as serum calcidiol levels have been associated with both a slightly higher and with a slightly lower risk of different skin cancers. The authors wrote that this may possibly be explained by the fact that serum calcidiol analyses do not provide information on the metabolism of vitamin D in the human skin, which can express enzymes that generate biologically active vitamin D metabolites or inactivate them.


    For this new study, researchers recruited 498 adult patients estimated to have an increased risk of skin cancer, including basal cell carcinoma, squamous cell carcinoma or melanoma, at the dermatological outpatient clinic of Kuopio University Hospital. Dermatologists at the University of Eastern Finland analyzed the patients’ background information and medical history and examined their skin. Patients were classified into different skin cancer risk classes: low, moderate, and high. Based on their use of oral vitamin D supplements, the patients were divided into three groups: non-users, occasional users and regular users. Serum calcidiol levels were analyzed in half of the patients and found to correspond to their self-reported use of vitamin D.


    A key finding of the study is that there were considerably fewer cases of melanoma among regular users of vitamin D than among non-users, and that the skin cancer risk classification of regular users was considerably better than non-users. Logistic regression analysis showed that the risk for melanoma among regular users was considerably reduced, more than halved, compared to non-users.


    The findings suggest that even occasional users of vitamin D may have a lower risk for melanoma than non-users. However, there was no statistically significant association between the use of vitamin D and the severity of photoaging, facial photoaging, actinic keratoses, nevus count, basal cell carcinoma and squamous cell carcinoma. Serum calcidiol levels were not significantly associated with these skin changes, either. Since the research design was cross-sectional, the researchers were unable to demonstrate a causal relationship.


    “…the question about the optimal dose of oral vitamin D in order to for it to have beneficial effects remains to be answered. Until we know more, national intake recommendations should be followed,” Professor of Dermatology and Allergology Ilkka Harvima of the University of Eastern Finland noted.

    Yingtao “Jerry” Zhao, PhD, an assistant professor of biomedical sciences at the College of Osteopathic Medicine (NYITCOM), now leads a three-year study thanks to a $428,400 grant from the National Institutes of Health (NIH), National institute of Neurological Disorders and Stroke (NIND). Targeting some of the most challenging neurological diseases from Alzheimer’s disease and Parkinson’s disease to autism spectrum disorders, the New York City based team seeks to improve an understanding of brain cell function and along the way develop new treatments targeting these unmet needs.


    As reported recently by NYITCOM’s Kim Tucker Camp, brain diseases and disorders adversely impact one in six persons worldwide. It turns out that the sugar molecule heparan sulfate links to some of these conditions. They cover the surface of all cells in the human body, and they may help self-regulate cell to cell interactions, also referred to as cell signaling. Deficient levels of heparan sulfate are associated with these brain diseases, including Alzheimer’s disease and Parkinson’s disease. Little is known about how this sugar molecule regulates brain function.


    The Study Context

    Thanks to the funding, Zhao and team will set up a study with a mouse model to investigate genetics, molecular biology, and other biomedical factors that may impact the role of these sugar molecules in the brains of adult mammals.


    What’s the Hypothesis?

    The study team hypothesizes if key brain cells called astrocytes associate with healthy heparan function, then this will regulate behavior and gene expression.


    Analyzing heparan sulfate activity in the brain cells of mice affected by various genetic and biomedical conditions, key conditions will include gene mutations associated with sugar molecule irregularities, artificially introduced nucleic acids (DNA or RNA), and reduced or elevated gene expression. Their findings could lead to the development of new heparan sulfate-based drugs for various brain conditions.


    As reported by Kim Tucker Camp, this study offers important research opportunities for New York Tech’s medical and undergraduate students with two NYITCOM students and two undergraduates which are selected from the college’s Advanced Research Core program.


    Other NYITCOM researchers involved include Weikang Cai, Ph.D., assistant professor of biomedical sciences; Yuan Huang, Ph.D., senior research associate; and Sohyun Moon, Ph.D., a postdoctoral fellow.

    Abstract

    The glomerular filtration barrier consists of podocytes, the glomerular basement membrane, and endothelial cells covered with a glycocalyx. Heparan sulphate (HS) in the glomerular filtration barrier is reduced in patients with proteinuria, which is associated with increased expression of the HS-degrading enzyme heparanase. Previously, we showed that heparanase is essential for the development of proteinuria in experimental diabetic nephropathy. Vitamin D supplementation reduces podocyte loss and proteinuria in vitro and in vivo. Therefore, we hypothesize that vitamin D reduces proteinuria by reducing glomerular heparanase. Adriamycin-exposed rats developed proteinuria and showed increased heparanase expression, which was reduced by 1,25-dihydroxyvitamin D3 (1,25-D3) treatment. In vitro, adriamycin increased heparanase mRNA in the podocyte, which could be corrected by 1,25-D3 treatment. In addition, 1,25-D3 treatment reduced transendothelial albumin passage after adriamycin stimulation. In line with these results, we showed direct binding of the vitamin D receptor to the heparanase promoter, and 1,25-D3 dose-dependently reduced heparanase promoter activity. Finally, 1,25-D3-deficient 25-hydroxy-1α-hydroxylase knockout mice developed proteinuria and showed increased heparanase, which was normalized by 1,25-D3 treatment. Our data suggest that the protective effect of vitamin D on the development of proteinuria is mediated by inhibiting heparanase expression in the podocyte.

    A first-of-its-kind antitrust action was filed Tuesday against the British Broadcasting Corporation (BBC), The Washington Post, Reuters, and Associated Press, seeking damages totaling millions of dollars based on the collusive censorship of online health and political news publishers who dared to question the narrative relating to vital matters such as COVID-19 pandemic and vaccine mandates.     Background

    The lawsuit reveals that by March 2020, a partnership was created called the Trusted News Initiative (TNI) between Big Tech and legacy media to exclude rival publishers from the dominant internet platforms. On Dec. 10, 2020, TNI agreed to focus on combatting the spread of harmful vaccine disinformation. This coordinated effort is, by definition, a classic unlawful “group boycott” to damage the ability of smaller publishers to compete or even survive. Members of the TNI include legacy media outlets the BBC, The Washington Post, Reuters, Financial Times, and Associated Press, along with social media and tech giants—Twitter, Meta (Facebook/Instagram), Microsoft/LinkedIn, and Google/YouTube. TNI members collectively hold 90% of the overall social media market, a 90% share of the social networking market, 75% of the video hosting market, and over 90% of the search-engine market.


    Antitrust Violations

    The complaint points to multiple examples of what plaintiffs’ state violates antitrust laws. “By their own admission, members of the ‘Trusted News Initiative’ (‘TNI’) agreed to work together, and have in fact, worked together, to exclude from the world’s dominant Internet platforms rival news publishers who engage in reporting that challenges and competes with TNI members’ reporting on certain issues relating to COVID-19 and U.S. politics. For example, TNI members deemed the following to be ‘misinformation’ that could not be published on the world’s dominant Internet platforms: (A) claims that COVID originated in a laboratory in Wuhan, China; (B) claims that the COVID vaccines do not prevent infection; (C) claims that vaccinated persons can transmit COVID to others; and (D) claims that compromising emails and videos were found on a laptop belonging to Hunter Biden. …The TNI did not only prevent Internet users from making these claims; it shut down online news publishers who reported that such claims were being made by potentially credible sources, such as scientists and physicians.” Dr. Ben Tapper, Dr. Joseph Mercola, founder of The Gateway Pundit Jim Hoft, independent journalists Ben Swann and Erin Elizabeth Finn, independent news outlets TrialSite News and Creative Destruction Media, and health-freedom activists Ty and Charlene Bollinger join Robert F.     Abstract

    Although the COVID-19 pandemic has persisted for over 2 years, reinfections with SARS-CoV-2 are not well understood. We use the electronic health record (EHR)-based study cohort from the National COVID Cohort Collaborative (N3C) as part of the NIH Researching COVID to Enhance Recovery (RECOVER) Initiative to characterize reinfection, understand development of Long COVID after reinfection, and compare severity of reinfection with initial infection. We validate previous findings of reinfection incidence (5.9%), the occurrence of most reinfections during the Omicron epoch, and evidence of multiple reinfections. We present novel findings that Long COVID diagnoses occur closer to the index date for infection or reinfection in the Omicron BA epoch. We report lower albumin levels leading up to reinfection and a statistically significant association of severity between first infection and reinfection (chi-squared value: 9446.2, p-value: 0) with a medium effect size (Cramer’s V: 0.18, DoF = 4).

    From censoring critical information that was true to a U.S. government policy proactively attacking and even discrediting doctors and other scientists that had differing opinions to suppressing the opinions of even ordinary Twitter users, the level, magnitude, and extent of the censorship regime would be expected in the previous Soviet Union or Communist China but not in the United States of America.


    TrialSite has been extensively censored and harassed online during this pandemic. We started as a pro-industry, pro-vaccine, pro-business media channel focusing on translating biomedical and health research into an understandable form with an emphasis on clinical research transparency and accessibility. We brought widespread awareness about ivermectin research to the United States early on during the pandemic and not because we had any bias toward the drug; we simply found it fascinating that so many people and researchers were looking into it in low-and-middle-income countries early on in the pandemic.


    On Facebook and YouTube, when we published that Slovakia had temporarily authorized the use of ivermectin (the first European country to do so), our content was censored, even though we include government documentation proving this was the truth. It didn’t matter. The same thing happened to us when we published via social networks the Uttar Pradesh story.


    In a state with over 230 million people, public health agencies, in an aggressive outreach campaign, brought education, testing, and home medicine kits, including ivermectin, to every village, town, and city across the region. The healthcare apparatus there heroically turned around the Delta-driven surge at the time. The Indian initiative was so successful that the World Health Organization actually issued a press release celebrating what health agencies in Uttar Pradesh accomplished. But evidencing true intent—which wasn’t (and isn’t good), they omitted one fact—the contents of the home medicine kit, which, of course, included ivermectin. This kind of brazen, blatant suppression of truth represents signs of authoritarianism.


    YouTube financially punished TrialSite with one strike after another. We had gained substantial momentum, and each and every time we were on the verge of breakout numbers, they (Google’s YouTube) found a way to censor and demonetize our efforts. They even censored our Ivermectin Peru documentary, which was about as objective as any such content can be.


    An independent media and technology platform, we at TrialSite cannot be easily directly censored. We don’t depend on social media, we have our own means of publishing and broadcasting objective, unbiased news. That’s how we brought awareness of Ivermectin to the West.


    That didn’t stop dark forces from coming after us. An outfit called NewsGuard contacted us twice, seemingly empowered by high-up forces, demanding with an air of entitlement to understand our ownership structure and a lot of other confidential information. They wanted us to take down certain content that they considered critical for the vaccine….or else….And they even have paid services if you want to get your credibility back.


    We politely told them that the content they didn’t appreciate was published as an opinion and not as the news. Not surprisingly, the opinion pieces they demanded to be taken down were critical of COVID-19 vaccine safety. We welcomed them to publish a counter opinion on the TrialSite on its self-author feature.


    But frankly, outfits such as NewsGuard could care less about objective news, or for that matter, real scientific dialogue or debate—they only care about compliance, adherence, and frankly, submission to a shadowy order that’s now emerging thanks to the Twitter files, some lawsuits and other myriad points of light representing an invigorating revolution in news and media.


    A cabal of industry, government, and academic center actors and the front-end mainstream media and social tech platforms undoubtedly tied together by a power and money agenda may have at one point had good intentions as well. After all, politicization impacted the pandemic response, and there were even elements within society that didn’t think SARS-CoV-2 was even a real virus. Sometimes the government has an emergency and must respond—we get it and support that wholeheartedly. Early on during the pandemic, TrialSite was like a public service, connecting hundreds of patients to clinical trials and many dozens of doctors to trial sites as they wanted to get involved and help the research enterprise during the pandemic.


    At some point during the pandemic, many players with well-intentioned energies and agendas, undoubtedly for a variety of reasons, slipped into a truly dark place, one where any critical views, even if accurate and based on science, were considered blasphemous. Now it's time to light the world back up!


    One of the most objective and unbiased news and information sources out there covering biomedical and health research, I had someone highly placed in the U.S. government tell me that TrialSite’s a special, important website along with our YouTube channel. When asked why, one person that’s a director in Health and Human Services told me, “TrialSite is the only media that reports on the good data associated with the COVID-19 vaccines and the problem data associated with these products. This kind of balance and objectivity is critically important. Don’t sell it.”


    This official went on to tell me that center- to liberal-leaning news sources struggle to critique vaccines at all while the media on the right just attacks these products now. As always, the truth is typically somewhere in between.
